Production of Keloid Animal Model

چکیده
Keloids and hypertrophic scars are basically an overabundance of fibrosis in cutaneous healing process. Unfortunately, keloids are only found in human beings. So far no successful animal model of keloid has been reported. Objectives: To produce an animal model of keloid. Main Outcome Measure: Animal model of keloid. Design: Prospective quasiexperimental trial. Setting: Department of Pathology and Experimental Research Laboratory, University of Health Sciences, Lahore, Pakistan. Duration: One year. Sample Size: Twelve albino animals i.e. three mice, three rats, three Guinea pigs, and three rabbits. Sampling Technique: Simple Convenience. Material and Methods: Three different techniques of keloid production were used. First animal of each species was given skin punch excision only; second was given Transforming Growth Factor Beta 1 injection only; and third was given Transforming Growth Factor Beta 1 injection followed by skin punch excision. Results were noted at four weeks. Results: Out of twelve animals, keloid was produced only in one rabbit (ear) which was given Transforming Growth Factor Beta 1 injection followed by skin punch excision. Conclusion: Keloid animal model can be produced in rabbit ear by giving Transforming Growth Factor Beta 1 injection followed by skin punch excision of ventral surface skin.

Deep and superficial keloid fibroblasts contribute differentially to tissue phenotype in a novel in vivo model of keloid scar.
BACKGROUND Keloids are thick fibrous scars that are refractory to treatment and unique to humans. The lack of keloid animal models has hampered development of effective therapies. The authors' goal was to develop an animal model of keloids using grafted engineered skin substitutes composed of keloid-derived cells. To demonstrate the model's utility, differences between deep and superficial kelo...
متن کاملLETTER TO THE EDITOR Abnormal Responses of Keloid Tissue to Wounding Identified Using In Vitro Model System
Dear Sir, Keloids are thick, raised scars that represent an extreme form of abnormal scarring. Unlike normal scars, keloids extend beyond the original wound margin and rarely regress; instead, they tend to proliferate indefinitely.1-4 These bulky scars can significantly impair function due to itching, pain, and decreased range of motion5 and can negatively impact psychosocial well-being and ove...
متن کاملHsp70 Knockdown by siRNA Decreased Collagen Production in Keloid Fibroblasts
PURPOSE There are currently no consistently effective treatments for the excessive collagen produced by keloid fibroblasts. Previously, we reported that heat shock protein 70 (Hsp70) is up-regulated in keloid fibroblasts and keloid tissue. We, therefore, investigated whether Hsp70 is related to excessive collagen production in keloid fibroblasts. MATERIALS AND METHODS We inhibited Hsp70 in ke...
متن کاملFibroblasts cocultured with keloid keratinocytes: normal fibroblasts secrete collagen in a keloidlike manner.
Keloid scars represent a pathological response to cutaneous injury, reflecting a new set point between synthesis and degradation biased toward extracellular matrix (ECM) collagen accumulation. Using a serum-free two-chamber coculture model, we recently demonstrated a significant increase in normal fibroblast proliferation when cocultured with keloid-derived keratinocytes. We hypothesized that s...
متن کاملComparing Logistic and Michaelis-Menten Multiphasic Models for Analysis of in vitro Gas Production Profiles of some Starchy Feedstuffs
Two multi-phasic models (logistic (LOG) and Michaelis-Menten (MM)) with three sub-curves were used to describe gas production kinetics of corn (CG), barley (BG), wheat (WG) and triticale (TG) grains. In each model sub curve, 1 describes the gas production caused by fermentation of the soluble fraction, gas production caused by fermentation of the non-soluble fraction is described in sub curve 2...
